    Hi, my question is:

    In free solution, how will the affinity of binding change if the ligand has 4 instead of 1 binding sites to the receptor. Assume that the receptor only has one binding sites.

    How will the first binding of the receptor to the ligand influency the affinity(k) of the second one? (therefore will k2 smaller than k1?)(assuming the receptor will be locked in place once binding)

    If the receptor is embeded in the membrane of Golgi apparatus, and its ligand is in the Golgi lumen. How will the first receptor binding influence the affinity of the second receptor to the ligand?
